Can i primarily access this via its usb 3.0 port? i'd like to have this plugged into my computer so i can use it as a video source drive.

No you cannot. The usb ports are for additional usb storage and transferring to and from the EX4100. You cannot connect it directly to your computer.

You can connect it to your computer via an Ethernet port -- not a USB 3.0 port. I remove the WD MyCloud from the router and directly connect it to my computer for my yearly backups. The data transfers are MUCH quicker when there is a direct connection

Actually YES. If you buy one of those USB to Ethernet and plug it directly to your computer you'll create a network and you can access it and it's very fast. I have it and I use it.
